Which burns more calories while bike riding? (Standing while you peddle or Sitting while you peddle)?

Or does it not make that much of a difference?

Answer
technically standing BUT that being said to "stand" while riding a bike USUALLY means that you are hitting a very very hard cycle (up hill or hard unstable surfaces) when you are on a stationary bike (if you have ever been to a spinning class) they UP the resistance on the bike when they stand b/c it makes peddaling harder...so thus you would use more energy and burn more calories

How does one stand up and ride their bike?

Practice on flat ground until you get used to it. Then, first and foremost, make sure the gear is not too easy, you want some substantial resistance to your down ward weight. Then slowly weight one leg. If it is too easy, then increase the gear so that it is harder. Once you have a gear that can offer resistance slowly alternate weighting one leg, then the other.... don't do it fast, just slowly until you get the resistance, the weighting and unweighting, and the balance shifting from one side to the other. After you start o get a rhythm then you can vary it. You will always need to be in a higher (harder) gear standing than sitting.

Standing when riding is used only for short bursts of speed, or to regain speed or apply maximum effort on hills. Occasionally, it is a good way to stretch muscles on a long ride.
